Il débute au cinéma à vingt ans, par une très brève apparition dans Un justicier dans la ville, un film d'action américain réalisé en 1974 par Michael Winner, et dont le rôle principal est joué par Charles Bronson. Il poursuit dans deux téléfilms, Wilma puis Flesh and Blood réalisé par Jud Taylor.

Washington directed and starred in the films Antwone Fisher (2002), The Great Debaters (2007), and Fences (2016). Washington made his Broadway debut in Checkmates (1988). He won the Tony Award for Best Actor in a Play for starring in the Broadway revival of August Wilson's play Fences in 2010.
Cerebral and meticulous in his film work, he made his debut as a director with Antwone Fisher (2002); he also directed The Great Debaters (2007) and Fences (2016). In 2010, Washington headlined The Book of Eli (2010), a post-Apocalyptic drama.
